Abstract Symptomatic presentation of patients with intra-cranial malignancy is frequently non-specific and differentiating such from those benign conditions challenging. Accordingly, GBM presents as a medical emergency more than any other common cancer, implying that effective strategies for rapid simple diagnostic stratification are urgently required. Here we describe the detection glial malignancies based on enrichment identification Circulating Glial Cells (CGCs) in peripheral blood. The first case controlled study included blood 189 participants split into discrete training test sets (145 GBM, 44 CNS conditions). second 40 cases malignancies, 22 conditions, 24 solid tumors brain metastases, 500 healthy individuals no prior diagnosis cancer current suspicion cancer. A third prospective was also carried out 68 radiologically evident intracranial space-occupying lesion suspected malignancies. Blood specimens were blindly evaluated CGCs through TruBlood assay to determine clinical sensitivity specificity. 98% specificity observed detecting them both studies. We detected differentiated CNS, samples, metastases. In study, 56 positive CGS, 12 negative CGCs. All CGC confirmed all conditions. present non-invasive approach presenting suspicious but neurocognitive symptoms hence early Moreover, immunocytochemistry facilitates differentiation metastatic epithelial cancers.
